Gewinnung von D-Mannitol mit rekombinanten Escherichia coli Stämmen

Ziel dieser Arbeit war die Konstruktion eines rekombinanten Escherichia coli
Stammes, der aus D-Fruktose bzw. D-Glukose D-Mannitol, ohne Nebenprodukte,
bildet. Es wurde im ersten Schritt die NAD-abhängige Mannitol-2-Dehydrogenase
des heterofermentativen Milchsäurebakteriums Leuconostoc pseudomesenteroides
ATCC12291 in E. coli exprimiert. Trotz hoher Enzymaktivität war dieser E. coli
Stamm nicht in der Lage, in Ganzzell-Biotransformationen D-Mannitol aus DFruktose
zu bilden Die Koexpression der NAD-abhängigen Formiat-Dehydrogenase
des methylotrophen Mycobacterium vaccae N10 zur Kofaktor-Regeneration führte,
trotz hoher Aktivitäten der beiden Enzyme, nur zur Bildung geringer Mengen D-Mannitol
aus D-Fruktose und Formiat.
Da E. coli normalerweise D-Fruktose über das Phosphoenolpyruvat-
Phosphotransferase System aufnimmt, ist dieser Zucker intrazellulär phosphoryliert
und damit kein Substrat für die Mannitol-2-Dehydrogenase. Dieses Problem wurde
durch die zusätzliche Expression des Glukose-Facilitators aus Zymomonas mobilis
überwunden, der D-Fruktose und D-Glukose durch erleichterte Diffusion ohne
Phosphorylierung dieser Zucker, transportiert. Dieser E. coli Stamm war in der Lage,
innerhalb von 18 Stunden bis zu 1 M D-Mannitol mit einer Ausbeute von 84 - 90
mol% und einer maximalen spezifischen Produktivität von 9,5 g x (gZTM x h)-1 aus
D-Fruktose zu bilden. Da dieser Stamm D-Glukose als Substrat zur D-Mannitol Bildung
nicht nutzen konnte, wurden sowohl Glukose-Isomerasen zum Reaktionsansatz
zugegeben, als auch die Glukose-Isomerase aus E. coli koexprimiert. Daraufhin
bildete E. coli D-Mannitol aus D-Glukose als einziger Kohlenstoff und Formiat als
Elektronendonor, ohne Beteiligung von phosphorylierten Zwischenstufen.
Aim of this work was the construction of recombinant Escherichia coli strains for the
formation of D-mannitol from D-fructose and D-glucose in whole-cell
biotransformations. First the NAD-dependent mannitol dehydrogenase from
Leuconostoc pseudomesenteroides ATCC12291 was expressed in E. coli. Despite of
its high enzyme activity this E. coli strain was not able to form D-mannitol from Dfructose
as resting cell suspension in a whole-cell biotransformation. The
coexpression of the NAD-dependent formate dehydrogenase of the methylotrophic
Mycobacterium vaccae N10 for the regeneration of NADH resulted in the formation of
a minor amounts of D-mannitol from D-fructose and formate, again regardless of high
enzyme activities in the respective strain.
Because D-fructose uptake in wildtype E. coli is catalysed by the PTS-system, this
sugar is phosphorylated and therefore not a substrate of the mannitol
dehydrogenase. This problem was overcome by the coexpression of the glucose
facilitator of Zymomonas mobilis for uptake of D-fructose without concomitant
phosphorylation. This strain was able to form D-mannitol up to a concentration of 1 M
in a reaction time of 18 hours with a yield YD-mannitol/D-fructose of 84 - 90 mol% from
D-fructose and formate as substrates. The maximum specific productivity was 9,5 [gDmannitol
x (gCDW x h)-1 ].
However, this E. coli strain was not able to form D-mannitol from D-glucose.
Therefore, glucose isomerases were supplemented to reaction mixture and the the
glucose isomerase from E. coli was coexpressed. In both cases this resulted in Dmannitol
formation solely from D-glucose as carbohydrate source and formate as
source of redox equivalents without the participation of phosphorylated
intermediates.
