Reaktionstechnische Untersuchungen zur präparativen enzymatischen Synthese und Aktivierung der Neuraminsäurederivate 3-Fluorneuraminsäure und Glycolylneuraminsäure

Für die Funktionen von Glycoconjugaten in physiologischen Vorgängen nehmen Neuraminsäure und
ihre Derivate wegen ihrer Polyfunktionalität und der exponierten, häufig terminalen Anordnung in den
Oligosaccharidstrukturen, eine zentrale Rolle ein. Die vorliegende Arbeit befasst sich mit der Entwicklung nd Charakterisierung von präparativen enzymatischen Routen zur Synthese und Aktivierung
zweier interessanter Derivate der Neuraminsäure, der 3-Fluorneuraminsäure und der Glycolylneuraminsäure.
Die Fluorneuraminsäure ist als künstliches Derivat der Neuraminsäure bemerkenswert, das eventuell
für die Untersuchung von Mechanismen und Wechselwirkungen, an denen Neuraminsäure beteiligt
ist, geeignet ist. Das natürliche 19F-Isotop des Fluorkerns eignet sich als NMR-Kern zur Untersuchung
mit Kernresonanztomographie, das künstliche, radioaktive 18F-Isotop für Untersuchungen mit PET.
Als Zielverbindung wurde 3-Fluorneuraminsäure (Neu5Ac3F) ausgewählt.
Glycolylneuraminsäure ist deshalb von besonderem Interesse, weil sie im Gegensatz zu
NeuAc fast ausschließlich bei Tieren, nicht aber beim Menschen vorkommt. Bei einem zu
98-99% gleichen Genom von Affen und Menschen ist dieser Unterschied die einzige,
bislang gut charakterisierte biochemische Differenz zwischen Affen und dem Menschen. Da
es keine reichhaltige natürliche Quelle und bislang kaum effektive Ansätze für eine technische
Synthese von NeuSGc gibt ist die Entwicklung einer Synthese zur Bereitstellung der für
Untersuchungen der Funktion von NeuSGc benötigten Mengen von großem Interesse. Für
die Synthese von NeuSGc wurde ein zwei-Enzym-System aus Epimerase und Aldolase
genutzt.
The function of glycoconjugates in physiological proceedings are basically determinate by Neuraminic
acid and its derivatives due to their polyfunctionality and exposed, mostly terminal position in the
oligosaccharide structures. The concern of this thesis is the development and characterization of
preparative enzymatic routes for the synthesis and activation of two interesting Neuraminic acid
derivatives, the 3-Fluoroneuraminic acid and Glycolylneuraminic acid.
Fluoroneuraminic acid is remarkable as a synthetic derivative of Neuraminic acid, which might be
usable for the investigation of interactions in which Neuraminic acid is involved. The natural
19F-isotope of the Fluorine nucleus is suitable as a NMR nucleus for the investigation via magnetic
resonance and the artificial radioactive 18F-isotope for investigations via PET.
3-Fluoroneuraminic acid (Neu5Ac3F) was chosen as the target substance.
Glycolylneuraminic acid is of special interest because it is only found in animals, but not in human
beings. With a 98-99% equal genome of apes and humans, this is by now the only thoroughly
characterized biochemical difference between apes and humans. Since there is no rich natural source
and nearly no effective promise for a technical synthesis of Neu5Gc the development of a synthesis
which grants the necessary quantity for the investigation of the function of Neu5Gc is of high consequence.
For the synthesis of Neu5Gc a two-enzyme system of Epimerase and Aldolase was used.
