In der vorliegenden Promotionsarbeit sollten aus D-Aminosäuren bestehende Peptide (DPeptide)
identifiziert werden, die mit hoher Spezifität an Aβ binden. Für D-Peptide wird im
Vergleich mit den jeweiligen L-Enantiomeren eine gesteigerte Protease-Resistenz und eine
geringere hnmunogenität erwartet (Schumacher et al., 1996).
Zur Identifizierung solcher D-Peptide wurde ein Screening einer Peptid-Bibliothek gegen
Aβl-42, bestehend aus D-Aminosäuren, durchgeführt . Dabei kam eine auf dem Bakteriophagen
M13 präsentierte, randomisierte 12mer Peptid-Bibliothek zum Einsatz. Nach vier Selektionsund
Amplifizierungsrunden wurden durch dieses Spiegelbild-Phagen-Display-Verfahren
verschiedene Peptide selektiert. Dabei trat die Aminosäuresequenz Q S H Y R H I S P A Q V
(Aminosäuren im Ein-Buchstaben-Code) in 20 von 39 selektierten Peptiden auf. In
Fluoreszenz-Spektroskopie-Titrations-Experimenten mit dem Spiegelbild dieses am
häufigsten auftretenden Peptids (D-Pepl) wurde eine Bindung an synthetisches Aβl-42,
bestehend aus L-Aminosäuren, nachgewiesen. Eine Abschätzung der Dissoziations-
Gleichgewichts-Konstante (KD) mittels nicht-linearer Regression ergab einen Wert im
mikromolaren Bereich.
Durch eine in-vitro-Markierung von Aβ-Ablagerungen in Gehirn-Gewebeschnitten von
Alzheimer-Patienten wurde mit Fluorescein markiertem D-Pepl eine Bindung dieses DPeptids
an natürlich vorkommendes Aβ nachgewiesen. Amyloid-Ablagerungen in
Gewebeschnitten von Patienten mit anderen Amyloid-Erkrankungen wurden durch D-Pepl
nicht markiert.
In Cytotoxizitätstests zeigte sich ein positiver Einfluss des selektierten D-Peptids auf die
Cytotoxizität von Aβ. Der Einfluss ist von der D-Pepl-Konzentration und dem Zeitpunkt der

Zugabe von D-Pepl zur Aβ-Lösung abhängig. Die Ergebnisse der durchgeführten
Cytotoxizitätstests konnten nicht in das Nukleations-Elongations-Modell zur in-vitro
Fibrillenbildung nach Lansbury et al. (bestehend aus zwei Schritten : Bildung von
Nukleationskeimen, Elongation von Fibrillen) (Jarrett et al., 1993 ; Jarrett und Lansbury,
1993; Harper und Lansbury, 1997) eingeordnet werden. Eine Interpretation aller Ergebnisse
der Cytotoxizitätstests war erst durch erweitertes kinetisches Modell zur in-vitro
Fibrillenbildung nach Harper et al. (Harper et al., 1999) möglich, in dem auch Protofilamente
berücksichtigt werden. Dabei wurde ein Einfluss von D-Pepl auf das Aggregationsverhalten
von Aβ sowie unterschiedliche Wirkmechanismen bezüglich der Toxizität von
Protofilamenten und Fibrillen vorausgesetzt.
A screening of a randomized 12mer peptide library presented an bacteriophage M13 for
binding to Aβl-42 consisting of D-amino acids as target was performed. After four rounds of
selection and amplification different peptides were selected. The amino acid sequence of
Q S H Y R H 1 S P A Q V (all amino acids in one-letter-code) was obtained in 20 of 39
selected peptides. The mirror image of this dominating peptide (D-peptide) binds to synthetic
Aβl-42 consisting of L-amino acids as shown by fluorescence spectroscopy titration
experiments . An estimation of the dissociation equilibrium constant (KD) with non-linear
regression produced a value in the micromolar range.
Binding of D-Pepl to natural Aβ was verified through in-vitro-targeting of Aβ precipitations
in tissue sections derived from patients that suffered from AD. Amyloid precipitations in
tissue sections of patients that suffered from other amyloidosis disorders were not marked by
D-Pepl .
In cytotoxicity tests, it has been demonstrated that the D-peptide has a positive effect an the
cytotoxicity of Aβ . The influence depends an the concentration of the D-peptide and the
fibril formation state of Aβ. The results of the cytotoxicity assays could not be explained
based an the nucleations-elongations-modell by Lansbury et al. (consisting of two steps :
building of a aggregation nucleus and elongation of fibrils) (Jarrett et al., 1993 ; Jarrett and
Lansbury, 1993; Harper and Lansbury, 1997). An interpretation of all results of the
cytotoxicity tests becomes possible by consideration of an extended model by Harper et al..
which adjusts protofilaments (Harper et al., 1999). Thereby an influence of D-Pepl to the
aggregation of A(3 as well as different mechanisms for toxicity of protofilaments and fibrils
was assumed.

Identifizierung und Charakterisierung eines spezifischen Liganden für das Alzheimer Amyloid-ß-Peptid (Aß)

Die Alzheimersche Demenz (AD) ist eine progressive, neurodegenerative Erkrankung, in
deren Pathogenese der Amyloid-Kaskaden-Hypothese" zufolge das Amyloid-β-Peptid (Aβ)
eine zentrale Rolle einnimmt. Dabei wird davon ausgegangen, dass durch die progressive
Akkumulierung von Aβ komplexe, multizelluläre Kaskaden initiiert werden, die neuritische
Schäden, Aktivierung von Mikroglia-Zellen und Astrozyten, neuronale Funktionsstörungen
und den Verlust von Synapsen bewirken. Basierend auf dieser Hypothese werden derzeitig
Diagnose- und Therapiestrategien erarbeitet, in deren Mittelpunkt die Erkennung von
Aβ oder/und Aβ Fibrillen stehen.
Alzheimer's Disease (AD) is a progressive neurodegenerative disorder. Extracellular amyloid
plaques in the brain are one of the main histopathological hallmarks of this disease . The
amyloid-cascade-hypothesis" assigns the amyloid-β-peptide (Aβ) a central role in the
pathogenesis of AD. lt is assumed that progressive accumulation of Aβ initiates a complex
multicellular cascade that includes neuritic dystrophy, microgliosis, astrocytosis, neuronal
dysfunction and loss of synapsis. The consequences ofthis multicellular cascade are impaired
amnestic and cognitive functions of patients suffered by AD. Based an this hypothesis,
diagnostic and therapeutic strategies targeting Aβ and Aβ fibrils are being pursued.
In the present dissertation peptides consisting of D-amino acids (D-peptides) should be
identified that bind to Aβ with high specifity. D-peptides are thought to be protease-resistant
and less immunogenic than the respective L-enantimer (Schumacher et al., 1996).
