TWD-Pflanzen zeigen einen pleiotropen Phänotyp, der sowohl in starkem Maße die Morphologie
als auch den Entwicklungszyklus der Pflanze betrifft . In Kombination mit Interaktionsstudien
zellulärer Partner kann die Struktur des TWD-Proteins wichtige Informationen über die Prozesse
liefern, die für die Pflanzenarchitektur wichtig sind.
In dieser Arbeit wurden zunächst Protokolle zur heterologen Expression des TWD-Proteins in
E.coli und zur Reinigung des rekombinanten Proteins entwickelt. Durch molekularbiologische
Methoden wurde dem TWD C-terminal ein Affinitätstag angefügt. Die Reinigung erfolgte
sequentiell über IMAC, Gelfiltrations- und lonenaustauschchromatographie . Sowohl Reinheit als
auch Menge an rekombinantem Protein reichten für eine eventuelle Kristallisation aus. Allerdings
war es nicht möglich, das Protein auf mehr als 2 mg/ml zu konzentrieren, was unterhalb des
üblichen Grenzwertes von 5 mg/ml liegt. Aus diesem Grund wurde mit zwei verkürzten
Konstrukten des TWD-Proteins, die von T.Kamphausen (Max-Planck Forschungsstelle für die
Enzymologie der Proteinfaltung, Halle/Saale) zur Verfügung gestellt wurden, weitergearbeitet:
TWD ohne Membrananker (TWD- MA) und der N-terminalen FKBP-Domäne (TWD12D) .
Unter Verwendung von Natriumformiat konnte TWD-MA und von Ammoniumsulfat und Ammoniumphosphat
als Fällungsmittel konnte TWD12D kristallisiert werden. Zu bemerken ist die sehr
lange Kristallisationszeit von 3 Monaten bis zu einem Jahr. Im Fall des TWD-MA konnten
Synchrotrondatensätze (ESRF, Grenoble, Frankreich) dreier nativer und dreier mit Schweratomen
derivatisierter Kristalle aufgenommen werden.
Erste Röntgenbeugungsaufnahmen eines TWD12D Kristalles zeigen eine sehr gute Diffraktion, die
bei Synchrotronstrahlung eine Auflösung von etwa 2 Ä haben wird.
Ziel der weiteren Arbeiten wird sein, die Struktur des TWD-MA zu bestimmen, Datensätze der
TWD12D Kristalle aufzunehmen und ebenfalls deren Struktur zu bestimmen. Des Weiteren gilt es
die Kristallisationbedingungen weiter zu verbessern, um die Kristallisationszeit herabzusenken .
Neben den röntgenkristallographischen Untersuchungen wurden auch NMR-spektroskopische
Experimente zur Bestimmung der dreidimensionalen Struktur durchgeführt. Zu diesem Zweck
wurde das TWD12D mit Isotopen markiert. Ein kompletter Satz an dreidimensionalen,
hochauflösenden NMR-Experimenten zur Resonanzzuordnung wurde durchgeführt und mit der
Zuordnung der Resonanzen begonnen.
Erste Ergebnisse beider Techniken deuten darauf hin, daß N-terminal die ersten 40 Aminosäuren
unstrukturiert sind. Daher ist es sowohl für die Qualität der Kristalle als auch für die Auswertung
der NMR-Experimente zu überlegen, mit einem um diese Aminosäuren verkürzten Protein zu
arbeiten.

important for the architecture of the plants.
In the present work, protocols for heterologous expression of the TWD-Protein in E.coli and
purification of the recombinant protein were developed . By using standard biomolecular techniques,
an affinity tag was fused to the C-terminus ofthe TWD-Protein. Sequential IMAC, gelfiltration, and
ion exchange chromatography was applied for purification . Although both purity and amount of
recombinant protein would have been sufficient for crystallization, it was not possible to
concentrate the protein to more than 2 mg/ml, which is sufficiently lower than the typical threshold
of 5 mg/ml. For that reason, all subsequent crystallization trials were conducted with two shortened
constructs of the TWD-Protein: TWD without membrane anchor (TWD-MA) and the N-terminal
FKBP-Domain (TWD12D), kindly provided by T.Kamphausen (Max Planck Reserach Unit for
Enzymology of Protein Folding, Halle/Saale) .
For succesfull crystallization ofTWD the presence ofthe precipitation agent sodiumformate was
crucial . In contrast, TWD12D did crystallize in the presence of either ammoniumsulfate or
ammoniumphosphate . In all cases the crystallization time was ranging from about 3 month to one
year. Native and heavy atom X-ray synchrotron datasets (ES-F, Grenoble, France) were collected
for TWD-MA.
First X-ray-diffraction patterns of the TWD12D-Crystals Show a good diffraction quality.
Resolution of about 2Ä can be expected for these crystals with X-ray synchrotron radiation .
Future goals are the calculation of the high resolution TWD-MA structure based an the available
data, and recording ofX-ray diffraction data an a synchrotron source forTWD12D followed by
structure calculation . Additional crystallization attempts will be directed at shortening the
crystallization time.
In addition to the X-ray crystallographic analysis NMR-based 3D-Structure determination of
TWD12D was initiated. TWD12D was labeled with NMR-active isotopes. A complete set of
standard threedimensional high resolution NMR-experiments for resonance assignment was
recorded and the assigment process was started .
Initial results from both techniques indicate, that the 40 N-terminal residues are not structured.
Therefore it is proposed to perform a complete structure analysis an a new construct, which is
lacking these 40 residues. Using such a shortened construct might aid both the crystallization
process and the NMR-Data analysis.
