Im Rahmen dieser Arbeit wurden mit Hilfe der Infrarot-Spektroskopie Untersuchungen
über den Zusammenhang zwischen der Struktur und den während des Funktionszyklus
ablaufenden Prozessen durchgeführt. Zu diesem Zweck wurden verschiedene
Strategien verfolgt:
Mit der vollreduzierten, CO gebundenen Rinderherzoxidase konnte erstmals gezeigt
werden, dass es möglich ist, mit einer Zeitauflösung von 5 µs Differenzspektren über
den Bereich von 2200 bis 950 cm-1 aufzunehmen. Die Analyse der Daten ergibt, dass
die transiente Bindung des fotolysierten CO an CUB sowie die Strukturänderung einer
etwa 12 A entfernten Aminosäure (E286) kinetisch gekoppelt sind. Diese Aminosäure
ist Teil eines vorgeschlagenen Protonenkanals und spielt eine entscheidende Rolle
beim Transport von Protonen ins Reaktionszentrum bzw. durch die Membran. Die
Ergebnisse lassen auf einen Mechanismus schließen, der den Eintritt des Sauerstoffs
unter Beteiligung der Aminosäure E286 kontrolliert.
Die Ergebnisse mit der halbreduzierten, CO gebundenen Cytochrom-c-Oxidase von
Rhodobacter sphaeroides zeigen erstmals, dass der Elektronentransfer vom Häm a3
zum Häm a an die Deprotonierung der oben schon beschriebenen Aminosäure E286
gekoppelt ist. Folglich ist nicht nur der K-Kanal am reduktiven Teil des Funktionszyklus
beteiligt, sondern auch der D-Kanal, was gängigen Modellen der Literatur
widerspricht.
Es konnte zweifelsfrei gezeigt werden, dass die Rekonstitution der bakteriellen
Cytochrom-c-Oxidase in Proteoliposomen keinen Einfluss auf das Protein hat; somit
konnte die ATR-Technik für weitere Experimente genutzt werden.
Die Auswertung von Redox-Differenzspektren des Wildtyps und der Mutanten,
aufgenommen mit der ATR-Technik, ergeben die präzise Zuordung eines
Bandenmusters im Differenzspektrum. Dieses Signal kann eindeutig der Umgebungsänderung
der Aminosäure E286 zugewiesen werden.
Zum ersten Mal konnte im IR ein Intermediatspektrum des oxidativen Teils des
Funktionszyklus aufgenommen werden. Die Interpretation der Differenzspektren
demonstriert, dass ein Tyrosin beim Übergang in den Zustand F involviert ist und
deprotoniert vorliegt.
In this work, Fourier-transform infrared spectroscopy was applied to provide insight
into the structural and functional relationships of the complex catalytic cycle. To
accomplish this, different strategies were applied:
The results obtained with the fully reduced, CO-bound cytochrome c oxidase from
bovine heart showed for the first time the possibility of performing difference
spectroscopy on this enzyme with a time-resolution of 5 µs in the range of 2200 to 950
cm-l. The evaluation of the data revealed a dynamic link between the transient
binding of CO to CUB and the movement of an amino acid side chain (E286)
approximately 12 A away. This amino acid is part of the D-channel and thought to play an
important role in guiding the protons either into the binuclear center or through the
membrane. The results presented in this work suggest a mechanism in which oxygen
binding is controlled via E286.
The outcome of the measurements with the two electron-reduced, CO-bound
cytochrome c oxidase from Rhodobacter sphaeroides indicate the deprotonation of E286
during electron transfer from heme a3 to heme a. This result contradicts the current
model of the catalytic-cycle because this deprotonation suggests an uptake of just one
proton rather than two through the K-channel.
Redox difference spectra of wild type and mutant cytochrome c oxidase obtained with
the attenuated total reflection technique pointed out the precise assignment of a band
feature. This signal was clearly attributed to a conforrnational change of the side
chain of amino acid E286.
For the flfSt time it was possible to record a difference spectrum in the IR of the
oxidative part of the catalytic cycle. The F minus O spectrum showed an involvement of
a tyrosine which undergoes deprotonation.

Die Cytochrom-c-Oxidase ist eine komplexe biologische Maschine, die einen
elektrochemischen Gradienten über einer Membran aufbaut. Getrieben wird dieser Prozess
durch die Reduktion von Sauerstoff zu Wasser. Neben den vier Protonen, die durch
die Membran gepumpt werden, müssen noch vier weitere Protonen für die
Sauerstoffchemie aufgenommen werden.
Cytochrome c oxidase is a complex biologica1 machinery , which couples the
reduction of oxygen to a vectorial transport of protons across the membrane. To achieve
this, the protein takes up eight protons, four electrons, and an oxygen molecule. The
electrons and four of the eight protons are used for the reduction of oxygen to produce
water. The remaining four protons are pumped across the membrane.
