In particular, we have investigated the reproducibility of the head coordinate system including
variability for different operators in order to estimate errors introduced by the head frame
definition. We further investigated the head loca1isation procedure and provide some esti-
mates about head movements during a typica1 MEG investigation. Thereby, we have devel-
oped strategies to improve both the variability of the head frame definition and the head loca-
tion accuracy. We will show that the use of a thermoplastic mask effectively reduces the inter-
operator variability and provides a head based coordinate system that is robust across different
operators. Wehave a1so introduced changes in the head location procedure where the im-
provements have reduced the head location error by a factor of more than 2 during rea1 meas-
urements.
At the end of the thesis we addresse the question of "how weIl can MEG loca1ise neuromag-
netic sources" by applying the weIl known equivalent current dipole (ECD) approach on
phantom data, where we have tried to simulate MEG measurements (with known sources)
under rea1istic conditions.

Die Magnetenzephalographie (MEG) ist eine nicht-invasive Methode zur Messung
schwacher Magnetfelder im Kopf, welches sich noch in der vorklinischen Phase
befindet. Das Verstehen und die Funktion des Gehirns, aber auch die Erforschung von
Gehirnkrankheiten (z. B. Alzheimer, Epilepsie) stehen im Mittelpunkt der
Neurowissenschaften. In der Forschung dient das MEG, wie auch die Elektroenzephalographie
(EEG), zur Detektion von neuronaler Aktivität mit einer sehr hohen zeitlichen
Auflösung. Der große Vorteil der MEG gegenüber der EEG ist, daß bei der MEG die
Leitfähigkeitsunterschiede im Kopf vernachlässigbar sind. Diese magnetischen
Felder haben ihren Ursprung in der gleichzeitigen Aktivierung von Tausenden oder gar
Millionen von Neuronen und sind extrem schwach, in der Größenordnung von 10-159
Tesla, was in etwa dem 109 -ten Teil des Erdmagnetfeldes entspricht. Erst die
Entwicklung einer Technik, bei der Meßspulen induktiv an sogenannte SQUIDs
(Superconducting QUantum Interference Devices) gekoppelt sind, hat die Messung solch
schwacher Felder ermöglicht. Um die Meßapparatur möglichst frei von Störungen zu
halten, stellt man sie in einen elektromagnetisch abgeschirmten Raum.
Das Ganzkopf-MEG-System, Magnes 2500 WH der Firma 4D-Neuroimaging, ist
1996 im Forschungzentrum Jülich installiert worden. Die Genauigkeit des Gerätes,
sowie das Zubehör und die bei der Benutzung auftretenden Fehler waren zu dem
Zeitpunkt bei diesem System noch nicht bestimmt worden.
Das Ziel der vorliegenden Arbeit ist, die Reproduzierbarkeit von Signalen als auch
die Genauigkeiten des Systems und der eingesetzten Methode zu überprüfen, und
mögliche Fehlerquellen zu vermeiden. Dazu sind eine Vielzahl von Experimenten
durchgeführt worden, um eine möglichst gute Abschätzung der Genauigkeiten
verschiedener Messungen die mit dem Magnes 2500 WH durchgeführt werden zu
treffen. Die daraus gewonnenen Ergebnisse sollen als Grundlage für weitere
Forschungsvorhaben dienen.
Magnetoencepha1ography is a non-invasive technique to study electrophysiologica1 processes
in the living human brain. These relatively new technique offers a very high tempora1 resolu-
tion, where the measured signals are rather independent of the differences of conductivity
within the human head. Modem whole head systems a1low the simultaneous measurement of
the magnetic flux at more than hundred locations outside the head.
However, the source loca1isation accuracy of the such systems highly depend on the accuracy
of the measurement procedure and the quality of the data. The thesis presented here addresses
questions relating to the loca1isation capability of magnetoencepha1ography (MEG), with em-
phasise on testing the accuracy of a typical whole head MEG system before any source recon-
struction is applied.
