Das neu entwickelte Hochtemperatur-Aufschlussverfahren (HTZ) verzichtet auf die
bisher übliche Fluorinierung und erlaubt einen relativ hohen Probendurchsatz. Dies wird
durch die Abtrennung sauerstoffhaltiger Verunreinigungen" der Probe und anschließendem
Aufschluss in einem Schritt erreicht . Dabei wird ein Mittelfrequenzgenerator
zur induktiven Aufheizung eines in eine doppelwandige Glasküvette
eingebrachten Reaktionsstabes aus Glaskohlenstoff genutzt. Der Reaktionsstab ist
oben mit einer Bohrung, die als Probenbehälter dient, versehen, welche ein Gemisch
aus Probenmaterial und Graphitpulver aufnehmen kann . Dieser Stab ist selbst von
einem dünnwandigen, oben geschlossenen Zylinder aus Glaskohlenstoff umgeben, um
einen Austausch des bei der Reaktion freiwerdenden, im CO gebundenen Sauerstoffs
mit dem Sauerstoff der Glasküvette zu verhindern. Nach Evakuierung der Küvette, kann
das Probenmaterial aufgeschlossen werden . Der Reaktionsstab wird zur Freisetzung
des Sauerstoffs aus biogenem Silikat bis auf eine Temperatur von ca. 1550°C erhitzt .
Dabei reagiert Graphit mit Siliziumdioxid zu Siliziumcarbid und Kohlenstoffmonoxid
(CO). Das entstandene Kohlenstoffmonoxid wird mit Hilfe flüssigen Stickstoffs unter
Verwendung eines Molekularsiebes (5Ä) in ein Probenröhrchen einkondensiert und
später in einem Isotopenverhältnismassenspektrometer (Doppeleinlasssystem) vermessen
. Zur Freisetzung des Sauerstoffs aus anderen Silikaten kann die induktive
Heizung bis auf Temperaturen von 2200°C erhöht werden . Die Temperatureichung
erfolgte mit Hilfe eines von der Physikalisch-Technischen Bundesanstalt geeichten
Quotientenpyrometers .
Die Ergebnisse der durchgeführten Versuchsreihen zeigen, dass biogene Silikate mit
dem neuen Verfahren vollständig und ohne Fraktionierung der Sauerstoffisotope in COMessgas
umgesetzt werden . Für biogene Silikate unterschiedlicher Herkunft sowie
einen internationalen anorganischen Standard konnten O-180-Werte mit einem
maximalen Messfehler von ±0,3‰ (maximale Abweichung vom Mittelwert) reproduziert
werden . Dies bedeutet im allgemeinen eine Standardabweichung von a≤5 ± 0,15‰. Die
Eichung des neuen Verfahrens mit einem internationalen Standard lieferte für
Materialien, deren O-180-Werte mit den bisherigen Verfahren publiziert vorliegen, eine
sehr gute Übereinstimmung.
Damit steht ein neuartiges Verfahren zur Bestimmung der Sauerstoffisotopenverhältnisse
biogener Silikate zur Verfügung. Es erlaubt einen wesentlich einfacheren
Zugang zu deren O-180-Werten und eröffnet damit eine Vielzahl neuer Anwendungsmöglichkeiten.
The newly developed technique is based an an extraction procedure using
temperatures of up to 2200°C which supersedes the commonly practised fluorination
with F2 or BrF5 in nickel tubes at 500 to 600°C. In addition it permits a relatively high
throughput of samples. The disintegration of the silica material and the separation of
oxygen containing molecules or molecular groups which are frequently attached to or
absorbed by the silica material is reached in one step. Basically, samples of silica are
introduced into a small borehole at the top of a rod made of glassy carbon . This rod is
housed in a glassy carbon cylinder, closed at the top which itself is enclosed by a
double-walled glass vessel made of quartz. After evacuation of the system, the glassy
carbon rod is inductively heated to the temperature needed for disintegration of the
particular material under investigation . If diatoms are to be analyzed which possess a
non crystalline structure, a temperature of about 1550°C is adequate . At this
temperature the Si02 will easily be decomposed, whereby the liberated oxygen is
converted to CO. Presently this CO is trapped in small glass tubes which contain a
molecular sieve (5Ä) kept at liquid nitrogen temperature . The carbon monoxide will
afterwards be measured off-line in a dual inlet isotope ratio mass spectrometer.
Temperatures are controlled by using a quotient pyrometer which was calibrated by the
Physikalisch-Technische Bundesanstalt which is the authority of the Federal Republic of
Germany for the field of metrology.
Test measurements demonstrated that the oxygen isotopes of biogenic silica can
quantitatively be liberated and converted to carbon monoxide. For this purpose biogenic
silica of different origins with different oxygen isotope ratios were measured, including
an international standard. The reproducibility in terms of O-180-values is in the order of
a≤5 ± 0,15‰ which is excellent in view of other methods currently in use.
This method introduces a new technique for the determination of the oxygen isotopes
from biogenic silica. It, thus, greatly simplifies research for palaeoclimate investigations
which necessitates oxygen isotopes as proxy signals .

Biogene Silikate und deren Sauerstoffisotopensignatur haben sowohl für die marine als
auch für die terrestrische paläoklimatologische Forschung aufgrund ihres ubiquitären
Vorkommens ein sehr großes Potenzial . Bedingt durch methodische Probleme bei
ihrem Aufschluss, sind die Sauerstoffisotope biogener Silikate jedoch bisher nur wenig
genutzt worden . Zielsetzung war daher die Entwicklung eines neuartigen, keine
Fraktionierungseffekte verursachenden Aufschlussverfahrens zur Extraktion der
Sauerstoffisotope.
The oxygen isotopes from biogenic silicates are extremely useful with regard to research
activities in marine and terrestrial palaeoclimatology. However, biogenic silicates
have so far only marginally been used as climate proxies because the quantitative
extraction of the stable oxygen isotopes is exceptionally difficult . The main problems
arise from two aspects: firstly, a satisfactory method is missing for the quantitative
liberation of the oxygen isotopes. Secondly, oxygen containing compounds or molecular
groups attached to or absorbed by the silicates are difficult to separate without
provoking isotope exchange reactions that alter the isotope ratio of a sample. The aim,
therefore, was to develop a new method by which the oxygen isotopes can
quantitatively be liberated from biogenic silicates . In addition any isotope shifts should
be prevented due to isotope exchange reactions with attached molecules or molecular
groups.
