Markierungsverfahren zur Synthese 4-[18F]Fluorprolyl-haltiger Peptide

Der Bedarf an selektiven Radiopharmaka für die PET -Diagnostik macht es erforderlich, neue
Markierungsmethoden zu entwickeln, um der stofflichen und strukturellen Vielfalt der zu
markierenden Verbindungen Rechnung zu tragen. Daher wurden im Rahmen dieser Arbeit an der
Stoffklasse der Peptide, die als Pharmaka zunehmend eine wichtige Rolle spielen, verschiedene
Konzepte zur 18F-Fluorierung mittels Prolylresten untersucht und bewertet.
Bei der direkten nukleophilen, Kryptat-gestützten 18F-Fluorierung des Modellpeptids Z-Pro-Leu-
Gly-4-(4R)-(TsO)Pro-OMe zeigte sich, daß das Peptid unter den basischen
Markierungsbedingungen nicht stabil und demzufolge eine 18F-Fluorierung nicht möglich ist.
Aufgrund der milden Reaktionsbedingungen, bei denen aliphatische Alkohole mit Perfluorbutan-
l-sulfonsäurefluorid fluoriert werden, bietet sich dieses Reagenz zur 18F-Fluorierung sensitiver,
Hydroxylgruppen besitzender Biomoleküle an. Durch Kryptat-katalysierte, nukleophile 18F-
Fluorierung von N,N-Bis(perfluorbutan-l-sulfonyl)anilin und durch Evaporation des
Aktivprodukts kann Perfluorbutan-l-sulfonsäure-[18F]fluorid in trägerarmer Form sowohl gelöst in Toluol
(RCA: 83 ± 3 %) als auch durch Adsorption auf einer Polystyrol-Matrix (LiChrolut® EN) mit
einer Gesamtausbeute von 75 ± 6 % erhalten werden. In Gegenwart einer äquimolaren Menge an
Perfluorbutan-l-sulfonsäurefluorid als Träger lassen sich N-Boc-Hyp-OMe mit einer
radiochemischen Ausbeute von > 90 % und die beiden Modellpeptide Z-Pro-Leu-Gly-Hyp-OMe und Z-Gly-
Leu-Hyp-Gly-Leu-OMe mit 40 - 45 % bzw. 30 - 35 % 18F-fluorieren, während bei der
trägerarmen Synthese lediglich ca. 1 % des 18F-Fluorproduktes erhalten wird. Der reaktionstechnisch
notwendige Trägerzusatz an Perfluorbutan-l-sulfonsäurefluorid erlaubt naturgemäß nur die
Darstellung von Radiotracern geringer molarer Aktivität.
Zur Durchführung der 18F-Fluorierung des Modellpeptids N-Boc-Hyp-Leu-OH, das über den C-
Terminus kovalent an ein Wang-Harz gebunden ist, mittels Perfluorbutan-l-sulfonsäure-
[18F]fluorid wird das 18F-Fluorierungsagens in Abwesenheit eines Lösungsmittels synthetisiert
und auf dem Wang-Harz adsorbiert. Die Ausbeute beträgt 31 ± 3 %. Die anschließend
durchgeführte 18F-Fluorierung des Peptids in Gegenwart einer äquimolaren Menge von Perfluorbutan-l-
sulfonsäurefluorid verläuft mit einer radiochemischen Ausbeute von lediglich 4 ± 1 %.
Als Alternative ließ sich die Kondensation von 4-[18F]Fluorprolin-methylester mittels TBTU an
den C-Terminus von Z-Pro-Leu-Gly-OH mit einer radiochemischen Ausbeute von 70 -80 %
unter trägerarmen Bedingungen durchführen. Der dazu notwendige 4-[18F]Fluorprolin-methylester
läßt sich durch 18F-Substitution an N-Boc-trans-4-(p-toluolsulfonyloxy)prolin-methylester, bei der
neben dem cis- jedoch auch das trans-Produkt entsteht, und anschließender
Boc-Gruppenabspaltung mit einer Gesamtausbeute von 30 - 40 % erhalten. Somit steht eine weitere prosthetische
Gruppe zur trägerarmen Markierung von Peptiden zur Verfügung.
The requirement of selective radiopharmaceuticals for PET -diagnostics necessitates the
development of new label1ing methods taking into account the substantial and structural diversity
of the compounds to be label1ed. Therefore different concepts for 18F-fluorination of peptides,
which are increasingly important as pharmaceuticals, via proline moieties were explored and
evaluated in this work.
The direct nucleophilic kryptate based 18F-fluorination of the model peptide Z-Pro-Leu-Gly-4-
(4R)-(TsO)Pro-OMe showed, that the peptide is not stable under the basic label1ing conditions,
and as a result the 18F-fluorination is not possible.
Perfluorbutane-l-sulfonyl[ 18F]fluoride is a possible reagent for 18F-fluorination of sensitive
hydroxy groups containing biomolecules due to the mild reaction conditions at which aliphatic
alcohols are fluorinated by this reagent. By kryptate catalysed, nucleophilic 18F-fluorination of
N,N-bis(perfluorobutane-1-sulfonyl)aniline and evaporation of the active product no-carrier-
added perfluorobutane-1-sulfonyl[18F]fluoride can be achieved either dissolved in toluene (RCY:
83 ± 3 % ) or adsorbed on a polystyrole-matrix (LiChrolut® EN) with a total yield of 75 ± 6 %. In
the presence of an equimolar amount of perfluorobutane-1-sulfonylfluoride as carrier N -Boc- Hyp-
OMe can be 18F-fluorinated with a radiochemical yield of > 90 % and the two model peptides Z-
Pro-Leu-Gly-Hyp-OMe and Z-Gly-Leu-Hyp-Gly-Leu-OMe with 40 - 45 % and 30 - 35 %, while
in the case of no-carrier-added synthesis only ca. 1 % of the 18F-fluoroproduct is obtained. The
necessary addition of perfluorobutane-1-sulfonylfluoride carrier allows natural1y only the
synthesis of radiotracers with low molar activity.
For performance of the 18F-fluorination via perfluorobutane-1-sulfonyl[18F]fluoride of the model
peptide N-Boc-Hyp-Leu-OH, which is covalently bound to a Wang resin via the C-terminus, the
18F-fluorination reagent is prepared in absence of a solvent and adsorbed on the Wang resin with a
yield of 31 ± 3 %, based on [18F]fluoride. The subsequent 18F-fluorination of the peptide, which
was done in the presence of an equimolar amount of perfluorobutane-1-sulfonylfluoride, proceeds
with a radiochemical yield of merely 4 ± 1 %.
Altematively, the condensation of 4-[18F]fluoroproline-methylester via TBTU at the C-terminus
of Z-Pro-Leu-Gly-OH could be carried out under no-carrrier-added conditions with a
radiochemical yield of 70 - 80 %. The required 4-[18F]fluoroproline-methylester could be
achieved with a radiochemical yield of 30 - 40 % by 18F-substitution of N-Boc-trans-4-(p-
toluenesulfonyloxy)-proline-methylester, by which the trans-product is also formed in addition to
the cis-product, and the cleavage of the Boc group. As a result of these studies another prosthetic
group is available for no-carrier-added label1ing of peptides.
